Paper from a research group in i3S wins the first edition of Prémio Cancro Gástrico IPO do Porto

The paper entitled "A panel of intestinal differentiation markers (CDX2, GPA33, and LI‑cadherin) identifies gastric cancer patients with favourable prognosis" from the Differentiation & Cancer group at i3S has won the first edition of "Prémio Cancro Gástrico IPO do Porto", an award for the best article in gastric cancer research published in the past 2 years assigned by IPO Porto. The award ceremony will take place at IPO Porto on May 31st, 2021.

O artigo intitulado "A panel of intestinal differentiation markers (CDX2, GPA33, and LI‑cadherin) identifies gastric cancer patients with favourable prognosis" do grupo Differentiation & Cancer, no i3S, foi galardoado com a primeira edição do "Prémio Cancro Gástrico IPO do Porto", um prémio para o melhor artigo em investigação em cancro gástrico publicado nos últimos 2 anos e atribuído pelo IPO do Porto. A cerimónia de entrega do prémio terá lugar no IPO do Porto, no próximo dia 31 de Maio.

Precise integration of inducible transcriptional elements (PrIITE) enables absolute control of gene expression

In this study, the authors combined the use of genome editing tools and last generation inducible systems in order to develop a cell model where the expression of CDX2 transcription factor can be tightly regulated. CDX2 is an intestinal differentiation marker whose expression is often altered in gastric and colon carcinomas. Its control was here shown to be useful to uncover novel putative downstream effector genes (such as GPA33 and LDLR), and therefore, to further clarify the role of CDX2 in intestinal cells.

CDX2 homeoprotein regulates ST6GalNAc-I in intestinal metaplasia

This study, performed by a group of investigators at IPATIMUP and published in Laboratory Investigation, identified the marker of intestinal differentiation CDX2 as a modulator of the glycoproteome in intestinal metaplasia, a precursor lesion of gastric cancer characterized by a transdifferentiation of the gastric mucosa into intestinal mucosa. The study showed that CDX2 transcription factor directly regulates ST6GalNAc-I gene in intestinal metaplasia lesions.

Este estudo, realizado por um grupo de investigadores do IPATIMUP e publicado na revista Laboratory Investigation, identificou o marcador de diferenciação intestinal CDX2 como um modulador do glicoproteoma na metaplasia intestinal, uma lesão precursora de cancro gástrico que se caracteriza por uma transdiferenciação da mucosa gástrica em mucosa intestinal. O estudo mostrou que o factor de transcrição CDX2 regula directamente o gene da ST6GalNAc-I em lesões de metaplasia intestinal.

Differentiation reprogramming in gastric intestinal metaplasia and dysplasia: role for SOX2 and CDX2

In this work we have assessed the expression profile of two transcription factors, SOX2 and CDX2 in intestinal metaplasia (IM) and dysplasia, two lesions that precede gastric cancer onset.  Our results provide a molecular basis to understand the heterogeneity of metaplastic lesions and favor the hypothesis that dysplasia arises from IM. These results have clinical implications since they advise a closer surveillance of patients presenting with IM.

Reprogramação da diferenciação na metaplasia intestinal e displasia gástrica: um papel para SOX2 e CDX2

Neste trabalho caracterizámos o padrão de expressão de dois fatores de transcrição, SOX2 e CDX2 na metaplasia intestinal (MI) e displasia gástricas, duas lesões que precedem o desenvolvimento do cancro gástrico. Os nossos resultados providenciam uma base molecular para perceber a heterogeneidade das lesões metaplásicas e favorecem a hipótese de que a displasia surge a partir da MI. Estes resultados têm implicações clinicas uma vez que aconselham uma vigilância mais apertada de pacientes com MI.

Nanopartículas de quitosano modificado diminuem a expressão celular de CDX2 e atravessam o muco gástrico

A metaplasia intestinal (MI) do estômago é uma condição pré-maligna associada a um risco aumentado de adenocarcinoma gástrico, uma das principais causas de morte por cancro a nível mundial. O CDX2, o principal mediador da diferenciação intestinal, auto-regula a sua expressão e está ligado ao seu promotor na MI.A hipótese deste mecanismo ser crucial para a manutenção do fenótipo intestinal, torna o CDX2 um potencial alvo terapêutico.
