A New In Vivo Tool to Evaluate Breast Cancer Stem Cell Activity – the CAM

We have established a in vivo limiting dilution assay (LDA) using the chick embryo chorioallantoic membrane (CAM), using a metastatic breast cancer cell model previously shown to be enriched for cancer stem cell (CSC) properties. This new tool was able to reproduce the same results found in mice, which is the gold standard method to evaluate CSC enrichment. Importantly, the chick model has several advantages, namely its easy handling, accessibility, rapid growth, lower costs and the absence of ethical and regulatory constraints.

Uma nova ferramenta experimental para avaliar a atividade estaminal de células de cancro da mama in vivo

Estabelecemos um ensaio de diluições limitantes utilizando a membrana corioalantóide do embrião da galinha (CAM), usando um modelo metastático de cancro da mama, no qual já foi demonstrado um enriquecimento em células com potencial estaminal. Com este novo ensaio  foi possível reproduzir os mesmos resultados encontrados no ratinho, o modelo  animal standard para a avaliação do enriquecimento estaminal em cancro.

O tempo e a qualidade de vida dos doentes oncológicos têm aumentado sensivelmente nos últimos anos com a introdução de melhores tratamentos sistémicos. Contudo, o aumento na sobrevida tem algumas consequências, nomeadamente no aumento da incidência de complicações metastáticas. Neste contexto, as metástases cerebrais são ainda consideradas a complicação neurológica mais frequente e temível, uma vez que são responsáveis por sintomas incapacitantes ou por morte precoce, estando bastante associadas a mau prognóstico.

Nucleolin overexpression in cancer stem cells enables targeted intracellular delivery of synergistic drug combination

Breast cancer stem cells (CSC) are believed to play a relevant role in tumor growth and relapse, metastization and active evasion to standard chemotherapy, thus representing an important tumor target. The objective of this work was to assess whether nucleolin was a receptor expressed in both breast cancer stem cells and non-stem cancer cells and whether it could be exploited to target nanoparticles-encapsulating drug combinations.
